Operation rules and routine maintenance of optical microscope
1、 Rules for use of optical microscopes
(1) During the experiment, the microscope should be placed on the table slightly to the left in front of the table, and the mirror base should be about 6-7 cm away from the edge of the table.
(2) Turn on the light source switch and adjust the light intensity to the appropriate size.
(3) Turn the objective lens converter so that the low power lens is facing the through hole on the stage. First, adjust the lens to about 1-2cm away from the stage, then look into the eyepiece with the left eye, and then adjust the height of the concentrator to adjust the aperture aperture aperture to the maximum, so that the light enters into the lens barrel through the concentrator. At this time, the field of vision is bright.
(4) Put the slide to be observed on the stage, so that the observed part of the slide is in the center of the light hole, and then clamp the slide with the specimen holder.
(5) Observe with low power lens (10x objective lens, 10x eyepiece). Before observation, turn the coarse focusing hand wheel to raise the stage and gradually approach the slide. It should be noted that the objective lens should not touch the glass slide to prevent the lens from crushing the glass slide. Then, the left eye is fixed in the eyepiece, while the right eye is not closed (it is necessary to form the habit of opening the eyes and observing with a microscope, so that the drawing can be viewed with the right eye while observing), and turn the coarse focusing hand wheel to slowly lower the stage, and the enlarged object image of the material in the slide can be seen soon.
(6) If the object image seen in the field of vision does not meet the experimental requirements (the object image deviates from the field of vision), the moving handle of the stage can be adjusted slowly. During adjustment, it should be noted that the moving direction of the slide is opposite to that of the object in the field of vision. If the object image is not clear, you can adjust the micro focusing hand wheel until the image is clear.
(7) If a high-power objective is used for further observation, the part of the object image that needs to be magnified and observed should be moved to the center of the field of vision before switching the high-power objective lens (when the low-power objective lens is converted to the high-power objective lens, the object image range in the field of vision is greatly reduced). Generally, the low power objective lens and the high-power objective lens are basically in the same focus. When the low-power objective lens is used to observe clearly, the object image can be seen by changing the high-power objective lens, but the object image is not necessarily very clear, so it can be adjusted by turning the micro focusing hand wheel.
(8) After converting the high-power objective lens and seeing the object image clearly, the size of aperture diaphragm or the height of concentrator can be adjusted according to the needs to make the light meet the requirements (generally, when the low-power objective lens is replaced by the high-power objective lens, the field of vision will be slightly dimmed, so it is necessary to adjust the light intensity).
(9) After observation, the lens of the objective lens should be removed from the light hole, and then the aperture diaphragm should be adjusted to the maximum, and then the stage should be slowly lowered to check whether there is any damage to the parts (special attention should be paid to whether the objective lens is stained with water or oil. If it is stained with water or oil, it should be cleaned with lens paper). After inspection and treatment, the container can be packed.
2、 Maintenance of optical microscope
(1) Must master and strictly carry out the use rules.
(2) When taking and delivering the microscope, be sure to hold the curved arm with one hand and hold the base with the other hand. The microscope should not be tilted to prevent the eyepiece from sliding out of the upper end of the barrel. Handle the microscope with care.
(3) During observation, the position of the microscope should not be moved.
(4) The optical part of the microscope can only be wiped with special lens wiping paper. It is not allowed to wipe with other objects or touch the lens with fingers, so as to prevent the lens from being stained with sweat.
(5) Keep the microscope dry and clean to avoid contamination of dust, water and chemical reagents.
(6) When switching the lens of the objective lens, do not move the lens of the animal lens, only turn the converter.
(7) Do not turn the focusing handwheel at will. When using the micro focusing knob, the force should be light, the rotation should be slow, and when it can"t be turned, do not turn hard.
(8) It is not allowed to dismantle the parts on the microscope at will, and it is strictly forbidden to dismantle the lens lens at will, so as to avoid damaging the screw mouth of the converter, or making the low and high power objective lens out of focus after the screw opening is loose.
(9) When using high-power objective lens, do not use the coarse focusing handwheel to adjust the focal length, so as to avoid the damage of objective lens and glass slide due to excessive moving distance.
(10) Before returning after use, it is necessary to check whether there is water or reagent on the lens of the objective lens. If so, wipe it clean, and wipe the stage clean. Then put the microscope in the box and pay attention to the lock box.
